Sciex QTRAP 7500 platform

Achieving Comprehensive and High-Sensitivity Quantitative Data in Biomedical Research

In biomedical studies involving large sample sets, such as those in clinical proteomics, a key factor in generating robust and informative quantitative datasets is the ability to achieve high sensitivity without compromising the efficiency and precision of quantitative analysis. The growing scope of research areas and diversity of measurement protocols require reliable quantification of an expanding range of analytes within increasingly complex biological matrices.

The SCIEX Triple Quad 7500 Q-Trap Ready system, newly implemented at the CDT-CARD Proteomics Laboratory, enables targeted quantification of peptides and other bioactive molecules across diverse biological matrices with enhanced precision. The system’s advanced Scheduled MRM algorithm supports the development of complex targeted methods, facilitating comprehensive protein panel analyses. When operated in microflow chromatography mode, the SCIEX 7500 system can incorporate up to 4,000 MRM transitions within a single method, maximizing both analytical throughput and quantitative depth.

Orbitrap Exploris 480 platform

Mass Spectrometry as the Foundation of Modern Proteomics

Mass spectrometry has become the cornerstone of up-to-date proteomics research, enabling detailed analyses of protein composition and post-translational modifications. High-resolution instruments are now indispensable for advancing the field. Innovative proteomic methodologies are driving research into the mechanisms underlying civilization diseases, a key focus area at CDT-CARD.

The Orbitrap Exploris 480 mass spectrometer (Thermo Scientific) meets the rigorous demands of state-of-the-art proteomic investigations. The instrument’s orbitrap analyzer offers unparalleled mass resolution, reaching up to 480,000, coupled with exceptional measurement speed. These features support the full spectrum of proteomic workflows, including classical techniques (DDA, PRM), and advanced methodologies (DIA, also known as Next-Generation Proteomics).

Equipped with a FAIMS Pro ion mobility analyzer, the Exploris 480 introduces an orthogonal dimension to mass spectrometry, enhancing analytical performance. The FAIMS Pro reduces background noise and matrix effects, significantly improving the selectivity and sensitivity of proteomic analyses.

timsUltra AIP platform

Deeper proteomics insights into single cell proteomic (SCP) studies

The integrated platform comprises the following instruments: cellenONE X1, the Evosep Neo pump, and the timsUltra AIP mass spectrometer. The cellenONE system enables the precise isolation of single cells, as well as microbiopsies obtained by laser microdissection, along with their automated preparation for subsequent stages of analysis, significantly enhancing the reproducibility and accuracy of results.

The Evosep Neo pump is responsible for efficient peptide separation, offering predefined, standardized Whisper Zoom methods specifically optimized for high-throughput peptide separations while maintaining high sensitivity and reproducibility. The timsUltra AIP mass spectrometer integrates an ion mobility (IM) analyzer with a conventional time-of-flight (TOF) mass analyzer. This configuration introduces an additional variable—collisional cross section (CCS)—thereby expanding the analytical capabilities of the acquired data and enabling its more comprehensive use in the downstream data analysis software.

Overall, the configuration of this new platform ensures highly sensitive peptide identification and stable protein quantification, even at extremely low abundance levels, such as those encountered in single cells or microbiopsies obtained with laser microdissection.
